Abstract
Polygalacturonase (PG) is a group of pectinases that possess hydrolytic functions and are capable
to cleave the a-1, 4 glycosidic bonds between the two non-esterified galacturonic acid residues in
pectin and pectic substances. These enzymes are used commercially, such as paper, textile,
cellulose, and food industries and have many important industrial applications. It is the most
important tool for the purification of galacturonic acid and degradation of pollutants. In the
present study substrates orange peel and sugar cane bagasse were used for the production of
galacturonic enzymes by Trichoderma hazrianum through Solid State Fermentation (SSF). The
highest yield of enzymes produced was 183.0U/mL. This highest yield was obtained at pH7
temperature 35°C, inoculums size 5mL, moisture level 70%, and days of incubation5 nitrogen
level 2g.The purified enzyme has been further selected for ammonium sulphate precipitation and
gel filtration chromatography. The purified enzyme has been characterized through enzymatic
kinetic for study of its characterization temperature 45°C, pH8 and Km0.25, Vmax0.05 values.
The immobilization was observed through chitosan. The immobilized enzyme showed the
highest activity at temperature 45°C and pH6. For industrial application its activity was observed
to decrease turbidity and increase fruit juice clarification.
